Mechanism of Column Chromatography
According to the column chromatography mechanism, two different phases involved in the column chromatography mechanism.
- Stationary Phase – it is defined as a solid material which must have better adsorption capability and meets the standard criteria such as:
– Stability and inertness of the particles
– should be colorless, readily, and inexpensive
– uniform shape and size of the particle
– should be perfect for separation of complex mixtures of different compounds
– must allow the free movement of the mobile phase.
- Mobile Phase – this is nothing but made up of solvents and have some criteria such as:
– Sample or sample compound can be subjected into the column
– should act as a developing agent or helps in the movement of components
– should act as an eluting agent.
